In vitro and in vivo Estrogenic Effects of Fluorotelomer Alcohols in Medaka (Oryzias latipes)

چکیده
The present study demonstrates the estrogenic effects of two fluorotelomer alcohols, such as 1H,1H,2H,2H-perfluorooctan-1-ol (6:2 FTOH) and 1H,1H,2H,2H-perfluorodecan-1-ol (8:2 FTOH), in medaka (Oryzias latipes). An in vitro yeast two-hybrid assay indicated a significant, dosedependent interaction between medaka estrogen receptor α (ERα) and coactivator TIF2 upon treatment with 6:2 FTOH or 8:2 FTOH. In contrast, no interaction with the ERα was observed upon treatment with perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA), perfluorononanoic acid (PFNA), perfluorododecanoic acid (PFDA) or perfluoroundecanoic acid (PFUnDA). Expression analysis of hepatic vitellogenin (VTG) protein showed estrogenic potentials with, 6:2 FTOH and 8:2 FTOH, indicative of the induction of VTG synthesis in the livers of male medaka. We also investigated mRNA expression levels of two ER subtypes and two VTGs in the livers of male medaka following exposure to FTOHs. Quantitative real-time PCR analyses revealed that hepatic ERα, VTG I, and VTG II mRNA responded rapidly to FTOHs such as 6:2 FTOH and 8:2 FTOH after 8-h exposure, whereas no effects of these compounds on ERβ mRNA transcription were observed. These results strongly suggest that certain FTOHs, such as 6:2 FTOH and 8:2 FTOH, induce hepatic VTG through activation of ERα in male medaka.

Bridging the Gap From Screening Assays to Estrogenic Effects in Fish: Potential Roles of Multiple Estrogen Receptor Subtypes
This study seeks to delineate the ligand interactions that drive biomarker induction in fish exposed to estrogenic pollutants and provide a case study on the capacity of human (h) estrogen receptor (ER)-based in vitro screening assays to predict estrogenic effects in aquatic species. Adult male Japanese medaka (Oryzias latipes) were exposed to solutions of singular steroidal estrogens or to the...
متن کاملEffects of bisphenol a on the reproduction of Japanese medaka (Oryzias latipes).
We elucidated the effects of bisphenol A (BPA) on the reproductive capacity (fecundity and fertility) and estrogenic response of adult Japanese medaka (Oryzias latipes) and studied the transgenerational effects (F, generation growth and sex) of this agent on the F1 offspring. Eight mating pairs of medaka were exposed to BPA for three weeks at a measured concentration of 837, 1,720, or 3,120 mic...
متن کاملComparison of in vitro and in vivo bioassays for estrogenicity in effluent from North American municipal wastewater facilities.
Attempts to better understand causal factors affecting estrogenicity in municipal wastewater have primarily focused on analytical evaluation of specific chemical estrogens and the use of estrogen receptor (ER) based in vitro assays. To compare analytical,in vitro, and in vivo assays for estrogenicity, wastewater from four New York and one Texas municipal wastewater facilities was evaluated for ...
متن کاملThe phytochemical lindleyin, isolated from Rhei rhizoma, mediates hormonal effects through estrogen receptors.
Some plant compounds or herb mixtures are popular alternatives to conventional therapies and contain organic compounds that bind to some nuclear receptors, such as the estrogen receptor (ER), to exert various biological effects. We studied the effect of various herbal extracts on ERalpha and ERbeta isoforms. One herbal extract, Rhei rhizoma (rhubarb), acts as an agonist to both ERalpha and ERbe...
متن کاملEstrogen-like response of perfluorooctyl iodide in male medaka (Oryzias latipes) based on hepatic vitellogenin induction.
Using the in vivo male medaka model, we investigated the estrogen-like response of perfluorooctyl iodide (PFOI) which is a potential source of perfluorinated carboxylic acids. Using real-time quantitative polymerase chain reaction, the expression levels of related estrogenic genes including estrogenic receptor α (ERα), ERβ, vitellogenin I (VTG I), and VTG II in the livers of male medaka exposed...
